In a pot, combine sugar and milk. Bring to a boil over medium heat.

Let it boil for 2 1/2 minutes.

Remove from heat, then stir in peanut butter and vanilla until smooth.

Notes
For best results, use creamy peanut butter. Store in an airtight container at room temperature for up to 1 week, or refrigerate for up to 2 weeks. You can also freeze slices for longer storage.
